This study was aimed to explore the comparative acidifying properties of 2‐hydroxy‐4‐(methylthio) butanoic acid (HMTBA) and a combination of DL‐methionine (DLM) and acidifier in male broiler production. A total of 480 1‐day‐old broiler chicks were randomly divided into four treatments: A (low HMTBA, 0.057% HMTBA); B (low acidifier, 0.05% DLM + 0.057% acidifier); C (high HMTBA, 0.284% HMTBA); and D (high acidifier, 0.25% DLM + 0.284% acidifier). At 21 d, growth performance, chyme pH, digestive enzyme activities, and intestinal microflora were measured. The pH of crop, gizzard, and ileum contents was higher in the HMTBA treatment group than in DLM + acidifier treatment group. Furthermore, acidifier supplementation promoted growth of butyrate‐producing bacteria such as Faecalibacterium, whereas high HMTBA (0.284%) inhibited the proliferation of acid‐producing bacteria including Roseburia and Collinsella. The chymotrypsin activity was lower in the HMTBA group than in the DLM + acidifier group. In contrast, high‐level HMTBA group showed higher average daily gain and average daily feed intake than the DLM + acidifier group. These results suggested that HMTBA work through different pathways with DLM plus acidifier.  相似文献

For successful breeding programs, it is important to quantify the useful period of a male's reproductive life and it is often done simply by measurement of semen quality. This information is lacking for Japanese quail so we tested whether there is a decline in ejaculate quality and sperm kinematics with age, and whether the decline varies among strains. Nine males (n = 9) from each of 5 strains (A, B, C, D and E) were subjected to 4 semen collections (n = 16 per male) at 8, 16, 26 and 36 weeks of age. Ejaculate volume, sperm concentration and total sperm per ejaculate were measured, and sperm kinematics were analysed using a Sperm Class Analyser (SCA®). There was a significant effect of age for ejaculate volume, total sperm per ejaculate and per cent medium sperm. The effect of the interaction between age and strain was significant for percent progressive motile sperm, percent rapid sperm, velocity curvilinear, velocity straight line, velocity average path, linearity, straightness and beat cross frequency. Ejaculate volume peaked at Week 26 in all strains, while peak values for sperm concentration and total sperm per ejaculate were observed at Week 16 for most strains. There were declines in percent motile sperm, progressive motile sperm and rapid sperm, and in velocity curvilinear velocity, velocity straight line and velocity average path, by Week 16 for most strains. Linearity declined by Week 26 in some strains, and all strains showed a significant decline in beat cross frequency by that age. In conclusion, the ability of CASA to detect age-related changes in sperm kinematics makes it a valuable tool for identifying the best males and thus improving quail flock fertility. It is essential that breeders understand that age affects both sperm production and sperm kinematics, and that the changes vary with strain.  相似文献

Obtaining high crop yields with limited water consumption requires optimal irrigation strategies based on comprehensive studies of the parameters of plant–environment interactions. Here, we used a structural equation modelling (SEM) to assess the relationships among input irrigation factors and moderate factors to find an optimum water use efficiency (WUE) response factor, for outdoor and greenhouse cultivation of eggplant (Solanum melongena). The input irrigation factors (including irrigation interval, water salinity and environment) and the moderate factors (evapotranspiration, soil salinity, plant parameters, fruit parameters and crop yield) were used in water cycle algorithm (WCA) and genetic algorithm (GA) methods to optimize the water use efficiency. The optimization process included finding the best combination of irrigation factors and optimized eggplant cultivation. The structural equation modelling results indicate that irrigation interval negatively affected water use efficiency with a more dominant effect on plant parameters. Water salinity negatively affected the water use efficiency with a more dominant effect on soil salinity, crop yield and fruit parameters. Low salinity water was more effective than full irrigation to optimize the water use efficiency. The water cycle algorithm revealed that for outdoor cultivation, the optimal range of irrigation interval was 2–5 days and water salinity in the range of 0.8–2.2 ds/m. These factors optimized evapotranspiration (346.23–738.19 mm), soil salinity (4.16–9.45 ds/m), fruit parameters (33.81–35.12 cm) and crop yield (1715.7–2190.8 g/plant), as well as increasing the water use efficiency (3.08–4.89 g/(plant-mm)). Both the water cycle algorithm and genetic algorithm yielded very close to optimal values. Two years of repeated experiments and the closeness of the optimal values using the algorithms confirmed that the optimal amounts are reliable.  相似文献

Metal toxicity and soil acidity affect plant growth. A hydroponic experiment was conducted to simulate effects of pH and copper concentration [Cu] on early growth and biomass production of high-yielding rice seedlings (Oryza sativa L. cv. ‘NIAB 6’). The rice seedlings were allowed to grow in Yoshida nutrient solution with 1 (control), 8, 16, and 32 μM Cu at buffered pH levels of 4.5, 5.0, and 5.5, respectively. Shoot and root growth, leaf chlorophyll content, and root lipid peroxidation and K+ leakage of 15-day-old rice seedlings were determined. Results show that [Cu] had significant adverse effects on rice seedlings. Shoot height and root length were reduced by 18%–60% and 7%–69%, respectively, compared with the control, and shoot and root weight decreased by 26%–56%, and 25%, respectively, in response to progressive increases in [Cu]. Similarly, leaf chlorophyll content decreased by 150%–245% compared with the control treatment. Rice seedling roots suffered from significant increases in lipid peroxidation followed by K+ leakage (> 8–23 times) in response to increasing [Cu]. Averaged across [Cu], seedling growth and biomass were greater at pH 5.5 than at other pH treatments. In some instances, increasing pH minimized the adverse effects of Cu at low concentrations on rice seedlings.  相似文献

ABSTRACT

The study demonstrated S. alfredii is an excellent cadmium (Cd)/zinc (Zn) hyperaccumulator as Cd and Zn concentrations in leaves reached 2,183 and 13,799 mg kg?1 DW, respectively. There was a significant increase in root morphological parameters induced by 50 and 500 μM Zn supplement; however, a sharp decrease in these parameters occurred when treated with 100 μM Cd +1000 μM Zn. The inhibited root dehydrogenase activity in 100 μM Cd treated plants was restored to control levels when supplemented with 500 μM Zn. Moderate Zn supplement did not produce significant changes in (malondialdehyde) MDA concentrations as compared with those treated with Cd alone. Variations of the antioxidative enzymes proved an ineffective role in coping with metal-stress in S. alfredii. Combined Cd and Zn treatment significantly enhanced ascorbic acid (AsA) and glutathione (GSH) contents in leaves of S. alfredii, as compared with those treated with Cd alone. Thus, Zn may rely on the involvement of GSH in detoxification and tolerance.  相似文献

Background: The seladin-1 (selective Alzheimer disease indicator-1), also known as DHCR24, is a gene found to be down-regulated in brain region affected by Alzheimer disease (AD). Whereas, hair follicle stem cells (HFSC), which are affected in with neurogenic potential, it might to hypothesize that this multipotent cell compartment is the predominant source of seladin-1. Our aim was to evaluate seladin-1 gene expression in hair follicle stem cells. Methods: In this study, bulge area of male Wistar rat HFSC were cultured and then characterized with Seladin-1 immunocytochemistry and flow cytometry on days 8 to 14. Next, 9-11-day cells were evaluated for seladin-1 gene expression by real-time PCR. Results: Our results indicated that expression of the seladin-1 gene (DHCR24) on days 9, 10, and 11 may contribute to the development of HFSC. However, the expression of this gene on day 11 was more than day 10 and on 10th day was more than day 9. Also, we assessed HFSC on day 14 and demonstrated these cells were positive for β-ш tubulin, and seladin-1 was not expressed in this day. Conclusion: HFSC express seladin-1 and this result demonstrates that these cells might be used to cell therapy for AD in future. Background: Multiple sclerosis (MS) is known as a progressive central nervous system inflammatory disease. Certain factors, such as interleukins, inflammatory cells, and oxidative stress are supposed to involve in MS etiology. Because of the important role of oxidative stress, antioxidant therapy for MS has received more attention. Although coenzyme Q10 (CoQ10) acts as an antioxidant, there is a lack of enough research on its effects on MS. Therefore, the present research was designed. Methods: C57BL/6 female adult mice (n = 30) were used in this study. The animals were randomly divided into trial and control groups. To induce MS, routine procedure for experimental autoimmune encephalomyelitis (EAE) was used, and scoring was performed based on clinical signs. By detecting score one, CoQ10 administration was started (10 mg/kg/three weeks). By using ELISA and real-time PCR, the brain levels of TNF-, IL-10, IL-4, and IL-12 were studied. Statistical tests were used to analyze the data and the P value less than 0.05 was considered to be significant. Results: Clinical symptoms in EAE animals were significantly decreased (P<0.05) as compared to control ones. In addition, the level of the TNF- was significantly decreased following CoQ10 administration versus IL-10. The ratio of TH1/TH2 interleukins in treated animals was significantly less than that in non-treated animals (P<0.01). Conclusion: Our findings showed that CoQ10 is capable of suppressing the inflammatory pathway of MS.
